ABSTRACT

@#Objective To analyze the epidemiological characteristics of hospital admission cases with chemical poisoning in Guangdong Province from 2016 to 2020. Methods Data of hospital admissions for chemical poisoning in Guangdong Province from 2016 to 2020 were collected through Guangdong Province Health Statistics Network Reporting System,and descriptive analysis was conducted. The seasonal characteristics of the number of hospitalized poisoning cases were analyzed by the concentration method,the seasonal index(SI)was calculated by the weighted annual ratio averaging method,and the spatial auto-correlation of regional poisoning trend was analyzed by the global and local spatial autocorrelation. Results From 2016 to 2020,the number of hospital admissions for chemical poisoning in Guangdong Province was 54 656,showing a general decreasing trend. The sex ratio(male to female)was 0.90 ∶1.00. The top three types of hospitalization rates were carbon monoxide poisoning,alcohol poisoning and organic solvent poisoning. The main poisoning groups were students and children, farmers,workers and unemployed people,accounting for 31.74%,18.53%,13.91% and 10.39%,respectively. The 74.37% of poisoning cases were cured or improved and discharged,and the case fatality rate was 0.48%. The top three hospitalization rates in age group of 0-<5 years were organic solvent,metal and carbon monoxide poisoning. The hospitalization rate of carbon monoxide poisoning ranked the first among all age groups of ≥5 years. The top three regions with the highest average annual hospitalization rate were Shaoguan City (25.14/105 ),Qingyuan City (17.04/105 ) and Meizhou City (16.09/105 ). Carbon monoxide poisoning had a strong seasonality(M=0.77),with high incidence months of January,February and December(SI were 3.60,3.08 and 2.48,respectively). The inpatients with chemical poisoning showed non-random distribution and spatial correlation(all P<0.01),with a high-high clustering among 13 districts and counties in northern Guangdong(all P<0.05). Conclusion The number of hospital admission cases of chemical poisoning showed an overall decreasing trend in Guangdong Province from 2016 to 2020. The main types of poisoning were carbon monoxide poisoning,alcohol poisoning and organic solvent poisoning. The spatial distribution of chemical poisoning types showed spatial correlation and there were high-high clustering areas.

ABSTRACT

Stolon is an important organ for reproduction and regeneration of Amana edulis. Previous analysis of transcriptome showed that MYB was one of the most active transcription factor families during the development of A. edulis stolon. In order to study the possible role of MYB transcription factors in stolon development, the authors screened out an up-regulated MYB gene named AeMYB4 was by analyzing the expression profile of MYB transcription factors. In the present study, sequence analysis demonstrated that AeMYB4 contained an open reading frame of 756 bp encoding 251 amino acids, and domain analysis revealed that the predicted amino acids sequence contained two highly conserved SANT domains and binding sites for cold stress factor CBF. By multiple sequence alignment and phylogenetic analysis, it is indicated that AeMYB4 clustered with AtMYB15 from Arabidopsis thaliana, belonging to subgroup S2 of R2 R3-MYB. And most of the transcription factors in this subfamily are related to low temperature stress. The GFP-AeMYB4 fusion protein expression vector for subcellular localization was constructed and transferred into Agrobacterium tumefaciens to infect the leaves of Nicotiana benthamiana, and the results showed the protein was located in the nucleus. To investigate the transcriptional activation, the constructed pGBKT7-AeMYB4 fusion expression vector was transferred into Y2 H Gold yeast cells, which proved that AeMYB4 was a transcription activator with strong transcriptional activity. Real-time quantitative PCR was used to detect the expression of AeMYB4 gene in three different development stages of stolon and in leaves, flowers, and bulbs of A. edulis, which indicated that AeMYB4 transcription factor was tissue-specific in expression, mainly in the stolon development stage, and that the expression was the most active in the middle stage of stolon development, suggesting that AeMYB4 gene may play an important role in stolon development. This study contributes to the further research on the function of AeMYB4 transcription factor in stolon development of A. edulis.

ABSTRACT

Objective@#To understand the pathogen spectrum of severe hand, foot and mouth disease (HFMD), and analyze the genetic characteristics of enterovirus A71(EV-A71) in Xianyang in 2018.@*Methods@#Totally 67 specimens of severe cases of HFMD were collected. Enteroviruses associated with HFMD were detected by real-time PCR and the genotypes of enteroviruses were identified by VP4 region of enteroviruses. The nucleotide and amino acid sequences of VP1 region of EV-A71 were analyzed.@*Results@#A total of 30 samples were positive for enterovirus among samples from 67 severe cases with HFMD, including 9 cases of EV-A71, 11 cases of coxsackievirus A6 (CV-A6), 5 cases of coxsackievirus A16 (CV-A16), 2 cases of coxsackievirus A10 (CV-A10) and 2 cases of coxsackievirus A4 (CV-A4). The nucleotide and amino acid homologies of EV-A71 among 4 strains reached 96.7%-99.9% and 99.3%-100% respectively. The 4 strains of EV-A71 belonged to C4a subtypes by phylogenetic analysis. The six amino acid composite model was KADSTV in 4 strains of EV-A71. The EF region and GH region in antigenic determinants of 4 strains of EV-A71 kept consistent with representative reference strains, however, the EV-A71 SZK222 and SZK497 strains developed mutation at site 93 (I93V) of BC loop region.@*Conclusions@#EV-A71 and CV-A6 are major agents of severe HFMD in Xianyang in 2018. The genotype of EV-A71 belonged to C4a subtype and the VP1 gene did not show more mutations.

ABSTRACT

Objective@#To conduct a characteristic analysis of the case of acute hydrogen sulfide poisoning in fishing boats for nearly ten years in a city, and to research on prevention and control measures, so as to provide scientific basis for formulating prevention strategies.@*Methods@#In July 2018, collecting the data of acute hydrogen sulfide poisoning in fishing boats recorded by the Oceanic and Fishery Department & Health Sector from January 1, 2009 to June 30, 2018, the accident characteristics of acute hydrogen sulfide poisoning in fishing boats was analyzed, and compared the differences of hydrogen sulfide poisoning under different classification statistics.@*Results@#In the past ten years, there were 14 acute poisonings in the city, 34 people were poisoned and 12 deaths (the case fatality rate was 35.3%) . According to the types of fishing vessels, the majority number of acute hydrogen sulfide poisoning incidents, poisoned persons and deaths occurred on fishing boats, accounted for 71.4% (10/14) , 76.5% (26/34) and 75.0% (9/12) respectively. From the location of the accident, 85.7% (12/14) of the acute hydrogen sulfide poisoning incidents occurred outside the fishing port. Fisher's exact test showed that the case fatality rate in within the fishing port was higher than that outside the fishing port (P=0.008) . From the cause of direct poisoning, the first three reasons were enter the fish room to rescue, enter the fish room to move fresh catch, to clean the fish room. The poisoning showed seasonal changes, Fisher's exact test showed that the fatality rate in summer was significantly higher than that in spring (P=0.044) , but there was no statistical significance in other seasons (P>0.05) .@*Conclusion@#The acute hydrogen sulfide poisoning of fishing boats in this city has a high case fatality rate, frequent collective poisoning, and showed seasonal changes. Blind rescue, incomplete facilities, and inadequate system are important factors leading to poisoning, so daily measures and rescue measures should be strengthened to reduce the occurrence of poisoning and casualties.

ABSTRACT

Objective: The molecular cloning and prokaryotic expression of the transcription factor AsWRKY62 of Aquilaria sinensis were carried out, at the same time, the bioinformatics analysis and expression pattern analysis were also performed. The purpose of this study was to lay a foundation for further study on the role of AsWRKY62 in the growth and development of A. sinensis and the formation of agarwood. Methods: With the cDNA isolated from A. sinensis callus as template, the full-length coding sequence (CDS) of AsWRKY62 was amplified using RT-PCR and PCR method. The recombinant vector pET-21a-AsWRKY62 which was built and verified by gene recombination technique was transformed into E. coli BL21 (DE3) for prokaryotic expression and purification. The characteristics of physiochemical properties, conserved domains and subcellular localization of AsWRKY62 were calculated by a series of bioinformatics tools. The analyses of multiple sequence alignment of amino acid and phylogenetic tree were performed using DNAMAN and MEGA 5.0, respectively. The gene expression pattern in different tissues was detected by RNA-seq data. Results: The full length CDS of AsWRKY62 (GenBank accession MH925301) was 1581 bp, encoding a 526-aa protein which belongs to WRKY group I. The optimized induction conditions of recombinant pET-21a-AsWRKY62 were 0.5 mmol/L IPTG at 37 ℃ for 4 h. According to the tissue-specific expression pattern analysis, the AsWRKY62 gene in A. sinensis is mainly expressed in roots and stems, followed by agarwood and flowers. Conclusion: Cloning, expression and characterization of the AsWRKY62 gene for the first time indicated that it may be related to the formation of agarwood, which provided a theoretical basis for further study of its biological function.

ABSTRACT

OBJECTIVE: To clone AsWRKY25, a transcription factor gene of Aquilaria sinensis, for bioinformatic analysis and tissue expression analysis, and express its protein in prokaryotic cells, thus to lay a foundation for further study of the biological function of AsWRKY25. METHODS: With the cDNA isolated from A. sinensis callus as template, the full-length coding sequence (CDS) of AsWRKY25 was amplified using PCR method. The recombinant vector pET-28a-AsWRKY25 was transformed into Escherichia coli BL21 (DE3) for prokaryotic expression. The physiochemical properties and bioinformatic characters of AsWRKY25 were calculated by a series of bioinformatics tools and softwares. The expression patterns in different tissues were detected by RT-PCR. RESULTS: The full-length coding sequence of AsWRKY25 transcription factor was cloned from the callus of A. sinensis. The CDS of AsWRKY25 was 1 728 bp in length and contained a 1 728 bp open reading frame (ORF) encoding 575 amino acids. The CDS sequence was codon-optimized and then ligated into the pET-28a expression vector. The optimal induction condition of recombinant pET-28a-AsWRKY25 was 1 mmol•L-1 IPTG at 37℃ for 6 h in E. coli BL21 (DE3). The result of tissue expression analysis showed that AsWRKY25 had the highest expression level in the agarwood layer and the lowest level in roots, stems and branches. CONCLUSION: The WRKY transcription factor AsWRKY25 is successfully cloned from A. sinensis and expressed in E. coil. It is speculated that AsWRKY25 may be involved in the wound-induced agarwood-formation process in A. sinensis.

ABSTRACT

Objective To provide a basis for further study and utilization of the functional gene family of Pht1 in Salviae miltiorrhizae, the Pht1 gene family members from S. miltiorrhizae genome should be explored, identified and functionally predicted. Methods By utilizing the bioinformatics methods, based on the genomic and NCBI database of S. miltiorrhiza, the sequences of Pht1 family candidate genes of S. miltiorrhiza were obtained firstly by screening, and then carried on the multiple sequence alignment, conserved structure lookup as well as phylogenetic tree construction. Finally, the characteristic analysis and biological function prediction were carried out. Results The results showed that 12 Pht1 candidate genes were extracted from the genome of Salvia miltiorrhiza, together with the reported Smpht1, there are 13 Pht1 members in total, their homology as high as 74.34%, and nine members of them could provide more reliable functional annotation. Among the gene family members, Smpht1 & Sm1, Sm4 & Sm6, Sm5 & Sm11, in which these three pairs may share similar function as well as coevolution relation between each other; Smpht1, Sm1, Sm3, Sm5, Sm7, and Sm11, all of the six members can be only expressed in the roots of S. miltiorrhiza. furthermore, except Sm7 induced by p-deficiency and Sm3 member unaffected by the phosphorus supply factor, the other four probably belong to the P-deficiency induced up-regulated expressive genotype, which play an important role in phosphorus absorption of S. miltiorrhiza; Moreover, both Sm7 and Sm16 are possible to expressed in the flower organs of S. miltiorrhiza and then participate in the regulation of phosphorus balance in the flowering stage of plants. In addition, Sm14 is likely to be a mycorrhizal-induced specific expressive gene with the function of phosphorus transfer in Pht1 family members. Conclusion The Pht1 gene family was screened and identified systematically from the genome of S. miltiorrhiza for the first time, in addition, the members with characteristic and biological function were found out, which will provide dependable background support and ideas reference for the further research on the role of Pht1 in the growth and development of S. miltiorrhiza.

ABSTRACT

Objective To explore facial characteristics of patients with different organ diseases; To provide some references for objective study on TCM inspection information. Methods Smart TCM-I type Life Information Analysis Systems was used to detect facial characteristics of 510 patients with five zang-organs diseases. 36 specific quantitative parameters including red ?, green (G), blue (B) and hue (H), saturation (S), value (V) of the face, forehead, eyes, cheeks, nose and chin were collected, and the Kruskal M-Wallis H and Nemenyi test were used for statistical analysis. Results Among the 510 patients with five zang-organs diseases, 96 patients belonged to lung system diseases, 105 heart system diseases, 101 liver system diseases, 107 spleen and stomach system diseases and 101 kidney system diseases. There was statistical significance in R, G, B, H, S, and V in forehead, eyes, cheeks and nose. Conclusion Facial characteristics can provide objective references for the facial division of five zang-organs diseases.

ABSTRACT

OBJECTIVE: Excessive worry about minor matters and a state in which this worry is experienced as uncontrollable are known to be key symptoms of generalized anxiety disorder (GAD). Given the importance of pathological worry in GAD, the need for psychometrically sound measures of this construct has increased. The purpose of this study was to investigate the usefulness of the Korean version of Penn State Worry Questionnaire (K-PSWQ) for screening GAD. METHODS: Two hundred and forty six patients were initially screened, from which 102 GAD patients and 118 patients with anxiety disorder not otherwise specified (anxiety disorder NOS) were finally enrolled. Patients were diagnosed by a structured clinical interview for the DSM-IV Axis I. We also enrolled 114 control subjects who had no medical or psychiatric history. Pathological worry in both patients and control subjects were assessed at baseline using the PSWQ and we estimated optimal cutoff score by the receiver operating characteristic (ROC) analysis. RESULTS: We found that in the first ROC analysis, a score of 53 could simultaneously optimize sensitivity and specificity in order to discriminate GAD patients from control subjects. From the second receiver operating characteristic analysis, when both sensitivity and specificity were optimized, we can suggest a score of 61 as being the cutoff for differentiating GAD patients from patients with anxiety disorder NOS. CONCLUSION: The Korean version of PSWQ is a useful method for screening GAD patients, although ethnic and cultural differences may affect the cutoff score of PSWQ for GAD.

ABSTRACT

Objective: The purpose of this study is to investigate the correlation between various transcranial Doppler (TCD) ultrasonography parameters and clinical vasospasm after aneurysmal subarachnoid hemorrhage (SAH). Methods: This study enrolled 40 patients presented with aneurysmal SAH between September 2006 and August 2007. We measured differences of mean blood flow velocity (BFVm), highest systolic blood flow velocity (BFVh), and Lindegaard ratio (LR) in the middle cerebral artery on TCD examination. These parameters were evaluated for correlation with clinical vasospasm by univariate analysis and the receiver operating characteristic analysis. Results: Twelve patients (30%) developed clinical vasospasm. The best TCD parameters for the detection of clinical vasospasm were revealed to be differences of BFVm, BFVh, and LR values between 1st TCD test and 3rd TCD (7 cm/s, 11.5 cm/s, 0.45 respectively). The positive predictive value of any one of three parameters was 60% and the negative predictive value was 100%. Conclusion: TCD is still considered a useful tool for screening clinical vasospasm. To confirm the predictive value of the above parameters, further prospective study will be needed.
